NSW Police said the woman was travelling between Moruya and Wallaga Lake when the crash occurred sometime after 11pm on Wednesday, June 5.
The female driver was trapped inside a Holden Trax SUV which was found overturned in Kianga Creek on Thursday, June 6.
NSW Police said she did not regain consciousness until 4.30pm on Thursday.
"The trapped woman regained consciousness and was able to alert a relative; however, was unable to provide a location," a Police spokesperson said.
Officers from South Coast Police District began a search operation to locate the woman.
CEO of the Westpac Life Saver Rescue Helicopter Stephen Leahy said the rescue chopper was called to assist in the search at about 5pm.
"We had a crew up off the ground ready to commence the search but Police said the woman had been located and a high-level medical response was required," Mr Leahy said.
NSW Police said the mobile phone system was used to identify a potential location.
The Police said the car had left the highway and crashed into water.
"Within 20-minutes, police attended the Princes Highway at Kianga and located the Holden down an embankment, overturned in Kianga Creek," a Police spokesperson said.
NSW Ambulance, Fire and Rescue NSW, Moruya State Emergency Services (SES), Narooma Volunteer Rescue Association (VRA) and the Dalmeny Rural Fire Service arrived at the scene.
"Narooma VRA Rescue Squad were assisted by Moruya SES to extricate the woman from the vehicle and she was taken to hospital by ambulance," a VRA spokesman said.
The woman was rescued from the vehicle and treated for suspected fractures and exposure before being airlifted to Canberra Hospital in a stable condition.
